    Posthuman Perspectives and Postdramatic Theatre: the Theory and Practice of Hybrid Ontology in Katie Mitchell’s The Waves

    Drawing connections between the theories of Hans-Thies Lehmann’s postdramatic theatre and N. Katherine Hayles’s presentation of posthuman ontology, where each responds to technologically-conditioned ways of knowing, experiencing, and being, this article interrogates changing models of theatrical forms and subjectivity. It argues that Katie Mitchell’s 2006-2007 National Theatre production of The Waves constructs a particular kind of posthuman subject who is materially instantiated and formed by emergent processes. This hybrid being replaces the dualistically conceived sovereign subject/hero of the liberal humanist model typical of traditional drama, and articulates a world built of flatter structures of mutuality.Partiendo de los paralelismos entre las teorías del teatro postdramático de Lehmann y la presentación de la ontología posthumana de Hayles, que responden a los condicionamientos tecnológicos en las formas de aprehender la experiencia, el ser y el conocimiento, este artículo interroga los modelos cambiantes de la forma teatral y la subjetividad. Se sugiere que la producción de The Waves por Katie Mitchell en el National Theatre, UK, durante la temporada 2006-2007, construye un sujeto posthumano peculiar que se forma y sustancia materialmente por medio de procesos emergentes.     The Importance of Realism, Character, and Genre : How Theatre Can Support the Creation of Likeable Sociable Robots

    Stage plays, theories of theatre, narrative studies, and robotics research can serve to identify, explore, and interrogate theatrical elements that support the effective performance of sociable humanoid robots. Theatre, including its parts of performance, aesthetics, character, and genre, can also reveal features of human–robot interaction key to creating humanoid robots that are likeable rather than uncanny. In particular, this can be achieved by relating Mori’s (1970/2012) concept of total appearance to realism. Realism is broader and more subtle in its workings than is generally recognised in its operationalization in studies that focus solely on appearance. For example, it is complicated by genre. A realistic character cast in a detective drama will convey different qualities and expectations than the same character in a dystopian drama or romantic comedy. The implications of realism and genre carry over into real life. As stage performances and robotics studies reveal, likeability depends on creating aesthetically coherent representations of character, where all the parts coalesce to produce a socially identifiable figure demonstrating predictable behaviour

    Beyond Character: A Post/Humanist Approach to Modern Theatre

    The thesis explores what it means to be human; specifically, what characters in drama and theatre reveal about what it means to be human. It also explores what it means to talk about dramatic character; specifically, what the human's various forms .reveal about dramatic character and how such forms interact with critical approaches to character. The thesis, thus, has a dual focus but the human and dramatic character are, in the context of my project, importantly, entwined and mutually enlightening. One of the aims of this thesis is to rehabilitate dramatic character. In doing so, it works to rehabilitate humanist subjectivity, too, albeit of a sort that is modified by hybrid structures of being and identity which are informed by posthumanist discourse. Such a structure, I argue, enables humans to be conceived simultaneously as creators and creations. I name this structure post/humanist. The first three chapters consist of theoretically engaged discussions which present the post/humanist framework underpinning this thesis's arguments for identity, subjectivity, and modern dramatic character. Chapter One claims it is a mistake to view the modern human being in exclusively liberal humanist terms and employs Donna Haraway's cyborg to reveal and argue for its indeterminate post/humanist form. Chapter Two makes the case for this thesis's alternative post/humanist account of modern subjectivity by revealing that the representation of liberal humanist subjectivity as the orthodox form of the modern period may have been overstated. Chapter Three argues for a post/humanist method of analysing dramatic character that conceives it as a structure of natural and cultural parts. Chapters Four, Five, and Six present case studies of the characters of Shakespeare's Hamlet (circa 1599-1601), August Strindberg's Miss Julie (1888), and Sarah Kane's Blasted (1995).     Utilisation of Mucin Glycans by the Human Gut Symbiont Ruminococcus gnavus Is Strain-Dependent

    Commensal bacteria often have an especially rich source of glycan-degrading enzymes which allow them to utilize undigested carbohydrates from the food or the host. The species Ruminococcus gnavus is present in the digestive tract of ≥90% of humans and has been implicated in gut-related diseases such as inflammatory bowel diseases (IBD). Here we analysed the ability of two R. gnavus human strains, E1 and ATCC 29149, to utilize host glycans. We showed that although both strains could assimilate mucin monosaccharides, only R. gnavus ATCC 29149 was able to grow on mucin as a sole carbon source. Comparative genomic analysis of the two R. gnavus strains highlighted potential clusters and glycoside hydrolases (GHs) responsible for the breakdown and utilization of mucin-derived glycans. Transcriptomic and functional activity assays confirmed the importance of specific GH33 sialidase, and GH29 and GH95 fucosidases in the mucin utilisation pathway. Notably, we uncovered a novel pathway by which R. gnavus ATCC 29149 utilises sialic acid from sialylated substrates. Our results also demonstrated the ability of R. gnavus ATCC 29149 to produce propanol and propionate as the end products of metabolism when grown on mucin and fucosylated glycans. These new findings provide molecular insights into the strain-specificity of R. gnavus adaptation to the gut environment advancing our understanding of the role of gut commensals in health and disease

    SARS-CoV-2 susceptibility and COVID-19 disease severity are associated with genetic variants affecting gene expression in a variety of tissues

    Variability in SARS-CoV-2 susceptibility and COVID-19 disease severity between individuals is partly due to genetic factors. Here, we identify 4 genomic loci with suggestive associations for SARS-CoV-2 susceptibility and 19 for COVID-19 disease severity. Four of these 23 loci likely have an ethnicity-specific component. Genome-wide association study (GWAS) signals in 11 loci colocalize with expression quantitative trait loci (eQTLs) associated with the expression of 20 genes in 62 tissues/cell types (range: 1:43 tissues/gene), including lung, brain, heart, muscle, and skin as well as the digestive system and immune system. We perform genetic fine mapping to compute 99% credible SNP sets, which identify 10 GWAS loci that have eight or fewer SNPs in the credible set, including three loci with one single likely causal SNP. Our study suggests that the diverse symptoms and disease severity of COVID-19 observed between individuals is associated with variants across the genome, affecting gene expression levels in a wide variety of tissue types

    Effects of Anacetrapib in Patients with Atherosclerotic Vascular Disease

    BACKGROUND: Patients with atherosclerotic vascular disease remain at high risk for cardiovascular events despite effective statin-based treatment of low-density lipoprotein (LDL) cholesterol levels. The inhibition of cholesteryl ester transfer protein (CETP) by anacetrapib reduces LDL cholesterol levels and increases high-density lipoprotein (HDL) cholesterol levels. However, trials of other CETP inhibitors have shown neutral or adverse effects on cardiovascular outcomes. METHODS: We conducted a randomized, double-blind, placebo-controlled trial involving 30,449 adults with atherosclerotic vascular disease who were receiving intensive atorvastatin therapy and who had a mean LDL cholesterol level of 61 mg per deciliter (1.58 mmol per liter), a mean non-HDL cholesterol level of 92 mg per deciliter (2.38 mmol per liter), and a mean HDL cholesterol level of 40 mg per deciliter (1.03 mmol per liter). The patients were assigned to receive either 100 mg of anacetrapib once daily (15,225 patients) or matching placebo (15,224 patients). The primary outcome was the first major coronary event, a composite of coronary death, myocardial infarction, or coronary revascularization. RESULTS: During the median follow-up period of 4.1 years, the primary outcome occurred in significantly fewer patients in the anacetrapib group than in the placebo group (1640 of 15,225 patients [10.8%] vs. 1803 of 15,224 patients [11.8%]; rate ratio, 0.91; 95% confidence interval, 0.85 to 0.97; P=0.004). The relative difference in risk was similar across multiple prespecified subgroups. At the trial midpoint, the mean level of HDL cholesterol was higher by 43 mg per deciliter (1.12 mmol per liter) in the anacetrapib group than in the placebo group (a relative difference of 104%), and the mean level of non-HDL cholesterol was lower by 17 mg per deciliter (0.44 mmol per liter), a relative difference of -18%. There were no significant between-group differences in the risk of death, cancer, or other serious adverse events. CONCLUSIONS: Among patients with atherosclerotic vascular disease who were receiving intensive statin therapy, the use of anacetrapib resulted in a lower incidence of major coronary events than the use of placebo.
